目的:研究中药炎克宁汤剂对输卵管阻塞性不孕模型大鼠输卵管上皮Bcl-2及EGFR mRNA表达的影响。方法:96只8w~12w Wistar雌性大鼠随机分为正常对照组、模型组、妇乐冲剂组、炎克宁低中高剂量组,每组16只。造模后20d开始给药,给药后15d、30d处死各组大鼠。采用原位杂交法检测输卵管上皮细胞Bcl-2及EGFR mRNA的表达。结果:正常对照组输卵管未见有Bcl-2 mRNA表达,妇乐冲剂组与模型组比较15d、30dBcl-2mRNA表达明显升高(P<0.05),EGFR mRNA表达明显下降(P<0.05);炎克宁低剂量组Bcl-2及EGFR mRNA表达与模型组比较无显著差异;炎克宁中剂量组Bcl-2及EGFR mRNA表达与妇乐冲剂组比较无显著差异,与模型组比较有显著性差异(P<0.05);炎克宁高剂量组Bcl-2及EGFR mRNA表达与其他各组比较均有显著差异(P<0.05)。结论:解毒除湿、化瘀止痛治疗输卵管阻塞性不孕模型大鼠,能抑制输卵管细胞凋亡,可能与调节Bcl-2和EGFR mRNA蛋白表达有关。
OBJECTIVE: To study the effect of Chinese medicine Yankangning Decoction on the expression of Bcl-2 and EGFR mRNA in fallopian tube epithelium in oviduct obstructive infertility model rats. Methods: Ninety-six female Wistar rats aged 8w-12w were randomly divided into normal control group, model group, Fule Lecong group, and Yankanning low-medium high-dose group, with 16 rats in each group. The administration was started 20 days after the modeling and the rats in each group were sacrificed 15 days and 30 days after the administration. The in situ hybridization was used to detect the expression of Bcl-2 and EGFR mRNA in the oviduct epithelial cells. Results: No expression of Bcl-2 mRNA was found in the fallopian tube of the normal control group, and the expression of 30dBcl-2 mRNA was significantly increased (P<0.05) and the expression of EGFR mRNA was significantly decreased in the Fule Granule group compared with the model group on day 15 (P<0.05). The expression of Bcl-2 and EGFR mRNA in the low-dose Krynine group was not significantly different from that in the model group; the expression of Bcl-2 and EGFR mRNA in the middle-dose group was not significantly different from that in the Fule Granule group, and it was significant compared with the model group. Difference (P<0.05); The high-dose Yankanning group Bcl-2 and EGFR mRNA expression compared with other groups were significantly different (P<0.05). CONCLUSION: Detoxification, dehumidification, and phlegm-resolving treatment for tubal obstructive infertility model rats can inhibit apoptosis of the oviduct cells, which may be related to the regulation of Bcl-2 and EGFR mRNA expression.
