目的以院内制剂温胆颗粒为研究对象,建立快速、简便、环保、样品用量少、便于实现自动化和易与色谱分析技术联用的中药复方制剂挥发性成分分析方法。方法运用顶空固相微萃取技术富集温胆颗粒中的挥发性成分,并运用气相色谱-质谱联用分析技术结合NIST谱库检索分析其成分类别及相对含量。结果共检测出163种挥发性的成分,鉴定出74个色谱峰,占检出挥发性成分总含量的96.35%。其中主要化学成分是芳樟醇、丁香酚甲醚、异丁香酚甲醚、β-细辛醚、欧细辛醚、桔利酮、丁烯基苯酞、洋川芎内酯A、反式藁本内酯等,结合文献确定其主要来源为复方中的生姜、石菖蒲、川芎等药材。结论运用HS-SPME-GC/MS技术可快速检测温胆颗粒中的挥发性成分,并可应用于相关复方挥发性成分的血清药化学和脑组织液药化学研究工作。
OBJECTIVE To study the preparation method of volatile components of traditional Chinese medicine preparation with the preparation of Wengan granules in the hospital, which is fast, simple, environmental friendly, with less sample amount, easy to automate and easy to combine with chromatographic analysis. Methods Headspace solid-phase microextraction was used to enrich the volatile components of Wengan Granules. The components and their relative contents were analyzed by gas chromatography-mass spectrometry coupled with NIST library. Results A total of 163 volatile components were detected and 74 peaks were identified, accounting for 96.35% of the total volatile components. The main chemical components are linalool, eugenol methyl ether, isoeugenol methyl ether, β-asarone, asarone, citral, butenylphthalide, This lactone, combined with the literature to determine its main source of compound ginger, Shichangpu, Chuanxiong and other herbs. Conclusion HS-SPME-GC / MS can be used to detect the volatile components of Wengan Granule rapidly, and can be applied to the research of serum chemistry and brain chemistry of compound volatile components.
