目的:通过对五种哺乳动物(人、牛、狗、兔、鼠)血清白蛋白化学及热稳定性的调查研究,并且对构象稳定性进行比较,以获得不同物种白蛋白的结构信息。 疗法:化学稳定性通过使用盐酸胍(Gdncl)来评价,并且用X线测量法和圆二色法(CD)进行监控。热稳定性用差示扫描测热仪(DSC)进行评价。结果:在人、牛和鼠的白蛋白中,当X 线测量法监控Gdncl诱导变性时,观察到两次碱基转换,尽管在狗和兔的白蛋白中,只观察到一次碱基转换,但至少表明有一次稳定
OBJECTIVE: To investigate the chemical and thermal stability of serum albumin in five mammalian species (human, cow, dog, rabbit and mouse) and to compare their conformational stability to obtain the structural information of albumin in different species. Therapies: The chemical stability is evaluated by the use of guanidine hydrochloride (Gdncl) and monitored by X-ray measurements and circular dichroism (CD). Thermal stability was evaluated using a differential scanning calorimeter (DSC). RESULTS: In human, bovine and mouse albumin, two base transitions were observed when X-ray monitoring Gdncl-induced degeneration, although only one base transition was observed in albumin of dogs and rabbits, But at least one is stable
