现知某些疾病状态与白细胞介素6(IL-6)异常增高有关,烧伤病人血清中IL-6水平也增高,并已明确内毒素与IL-6的因果关系,但有关高浓度IL-6在导致免疫功能失调上的作用尚未明瞭。本文研究IL-6在体外对多种免疫指标的作用。选用5~8周龄的雄性C57BI/6小鼠,制成脾单个核细胞悬液,加激活剂或免疫调节剂培养72小时后,用3HTdR渗入法测定细胞的增殖情况。用生物素化抗IL-2R抗体与FITC-链霉亲和素偶合物作为试剂,以流式细胞仪测定IL-2R。分别以CTLL-16和B_0为靶细胞,以生物学方法测定IL-2和IL-6量。结果表明:(1)超过100u/ml的IL-6可抑制鼠脾淋巴细胞对刀豆素A(ConcanavalinA,ConA)的应答,在100~4000u/ml内,IL-6抑制作用呈剂量依赖性,最大抑制率为对照值的40~60%。高浓度IL-6对B细胞的LPS增殖应答无抑制作用,提示IL-6对T
It is known that certain disease states are associated with abnormally elevated interleukin-6 (IL-6) levels and increased serum levels of IL-6 in burn patients, and the causal relationship between endotoxin and IL-6 has been clarified. However, high concentrations of IL- 6 in causing immune dysfunction has not yet been clarified. This article studies IL-6 in vitro on a variety of immune indicators. Male C57BI / 6 mice aged 5-8 weeks were selected and spleen mononuclear cell suspension was prepared. After 72 hours incubation with activator or immunomodulator, the cell proliferation was measured by 3HTdR infiltration method. IL-2R was measured by flow cytometry using biotinylated anti-IL-2R antibody and FITC-streptavidin conjugate as reagents. CTLL-16 and B_0 were used as target cells to determine the amount of IL-2 and IL-6 by biological methods. The results showed that: (1) IL-6 over 100u / ml inhibited the response of mouse splenic lymphocytes to Concanavalin A (ConA).
