目的 研究端粒酶hTRT基因在睾丸肿瘤组织的表达及其意义。方法 应用核酸原位杂交技术对 5 1例男性睾丸肿瘤组织和 10例正常睾丸组织中端粒酶hTRT基因的表达进行检测和定位 ,并应用HPIAS 10 0 0高清晰度图像处理系统对hTRT阳性信号进行图像分析。结果 端粒酶hTRT基因在睾丸肿瘤组织中有极高的表达 ,其阳性率为 92 .16 % ( 4 7/5 1) ,端粒酶hTRT基因表达强度与肿瘤细胞的分化程度显著相关 ,睾丸良性肿瘤与睾丸恶性肿瘤间差异有显著性(P <0 .0 5 ) ,在睾丸肿瘤组织和癌旁组织及正常睾丸组织中的表达强度差异有非常显著性(P <0 .0 1) ,并且其强阳性表达水平与肿瘤细胞的分布定位一致。结论 端粒酶hTRT有可能成为睾丸肿瘤诊断的新标志物及治疗的新靶点。
Objective To study the expression of telomerase hTRT gene in testicular tumor and its significance. Methods The expression of telomerase hTRT gene in 51 male testicular tumor tissues and 10 normal testicular tissues was detected by nucleic acid in situ hybridization. The hTRT positive signal was detected by HPIAS10 0 high definition image processing system Perform image analysis. Results The telomerase hTRT gene was highly expressed in the testicular tumor tissues. The positive rate of telomerase hTRT gene was 92.16% (41/51). The telomerase hTRT gene expression intensity was significantly correlated with the differentiation of tumor cells. There was significant difference between benign and malignant testicular tumors (P <0.05), and there was a significant difference in the expression intensity between testicular tumor tissue and adjacent normal tissue and normal testicular tissue (P <0.01) And its strong positive expression level is consistent with the distribution of tumor cells. Conclusion Telomerase hTRT may be a new marker for the diagnosis of testicular tumors and a new therapeutic target.
