目的研究黄芩苷对卡氏肺孢子虫肺炎(PCP)大鼠血清肿瘤坏死因子-a(TNF-α)和白细胞介素-8(IL-8)含量的影响,以探讨中药黄芩苷对PCP大鼠的免疫调节作用。方法给SD大鼠皮下注射地塞米松磷酸钠建立PCP动物模型,观察该药对PCP模型建立前后肺组织病理及血清肿瘤坏死因子-α和IL-8含量的影响。结果大鼠血清TNF-α含量,黄芩苷预防组(2.14±0.14)及(2.57±0.15),(1.46±0.14),(1.12±0.13)ng/mL剂量组均高于正常对照组(0.70±0.21)ng/mL(P<0.05,P<0.01),低于模型对照组(3.65±0.73)ng/mL(P<0.01)。有效药物对照组(1.59±0.14)ng/mL明显高于正常对照组(P<0.01),低于模型对照组(P<0.01)。血清IL-8含量,预防组(0.43±0.11)ng/mL及低(0.47±0.10)ng/mL、中(0.39±0.11)ng/mL、高(0.32±0.11)ng/mL剂量组均低于模型对照组(0.65±0.12)ng/mL(P<0.01)。有效药物对照组(0.33±0.12)ng/mL低于模型对照组(P<0.01)。治疗组肺组织炎症较模型对照组明显减轻,肺泡腔泡沫样渗出物均明显减少。结论黄芩苷对PCP大鼠具有一定的免疫调节作用,能够降低TNF-α和IL-8含量,从而减轻炎症反应。
Objective To study the effect of baicalin on serum tumor necrosis factor-a (TNF-α) and interleukin-8 (IL-8) levels in rats with Pneumocystis carinii pneumonia (PCP) to explore the effects of baicalin on PCP Rat immune regulation. Methods The animal model of PCP was established by subcutaneous injection of dexamethasone sodium phosphate in SD rats. The effects of the drug on the pathological changes of lung tissue and serum tumor necrosis factor-α and IL-8 levels before and after the establishment of PCP model were observed. Results The content of serum TNF-α in rats was higher in the baicalin-prevented (2.14±0.14) and (2.57±0.15), (1.46±0.14), and (1.12±0.13) ng/mL dose groups than in the normal control group (0.70±). 0.21) ng/mL (P<0.05, P<0.01), lower than the model control group (3.65±0.73) ng/mL (P<0.01). The effective drug control group (1.59±0.14) ng/mL was significantly higher than the normal control group (P<0.01) and lower than the model control group (P<0.01). Serum IL-8 levels were low in the prophylactic (0.43±0.11) ng/mL and low (0.47±0.10) ng/mL, middle (0.39±0.11) ng/mL, and high (0.32±0.11) ng/mL dose groups. In the model control group (0.65 ± 0.12) ng/mL (P <0.01). The effective drug control group (0.33±0.12) ng/mL was lower than the model control group (P<0.01). The inflammation of the lung tissue in the treatment group was significantly less than that in the model control group, and the foam-like exudates in the alveolar space were significantly reduced. Conclusion Baicalin has a certain immunomodulatory effect on PCP rats, which can reduce the content of TNF-α and IL-8 and reduce the inflammatory response.
