目的探讨蓝莓对四氯化碳(CCl4)所致急性肝损伤大鼠肝组织核转录相关因子2(Nrf2)和Ⅰ型血红素氧化酶(HO-1)mRNA表达的影响。方法 SD大鼠48只随机分为正常对照组10只、蓝莓汁低、高剂量各组10只(1g/100g、2g/100g)、齐墩果酸(7.5mg/100g)组及模型组各9只,用CCl4致大鼠急性肝损伤模型。采用TRIzol提取肝组织总RNA,实时荧光定量PCR技术测定大鼠肝组织Nrf2及HO-1 mRNA的表达。结果与正常对照组比较,模型组肝组织Nrf2、HO-1 mRNA表达有所增加,差异无统计学意义。与模型组比较,蓝莓汁低、高剂量组及齐墩果酸组肝组织Nrf2、HO-1 mRNA表达明显增高,P<0.05。结论蓝莓可激活大鼠肝脏Nrf2增加HO-1的表达,提高机体的抗氧化应激能力,这可能是蓝莓预防CCl所致大鼠急性肝损伤的机制之一。
Objective To investigate the effect of blueberry on mRNA expression of nuclear transcription 2 (Nrf2) and type 1 heme oxygenase (HO-1) in liver of acute liver injury induced by carbon tetrachloride (CCl4) in rats. Methods 48 Sprague - Dawley rats were randomly divided into normal control group (n = 10), low and high - dose blueberry juice (10g / 100g and 2g / 100g), oleanolic acid (7.5mg / 100g) and model group 9 rats with CCl4-induced acute liver injury model. The total RNA of liver tissue was extracted by TRIzol, and the expression of Nrf2 and HO-1 mRNA in rat liver were detected by real-time fluorescence quantitative PCR. Results Compared with the normal control group, the expression of Nrf2 and HO-1 mRNA in liver tissue of model group increased, but the difference was not statistically significant. Compared with the model group, the expression of Nrf2 and HO-1 mRNA in the liver tissue of low, high dose and oleanolic acid groups were significantly increased (P <0.05). Conclusion Blueberry can activate Nrf2 in rat liver to increase the expression of HO-1 and increase the antioxidant capacity of the body, which may be one of the mechanisms of blueberry in preventing CCl-induced acute liver injury in rats.
