目的 :研究雌激素对人涎腺黏液表皮样癌血管生成的影响 .方法 :应用免疫组化、裸鼠肺移植瘤试验的方法探讨1 7β 雌二醇对人涎腺黏液表皮样癌高转移细胞系Mc3细胞的血管内皮生长因子的表达和肺移植成瘤能力影响 ;并应用免疫组化的方法对Mc3细胞的雌激素受体进行检测 .结果 :在体外 ,0 .1 μmol/L 1 7β 雌二醇可促进Mc3细胞血管内皮生长因子的表达 .在裸鼠体内实验中给药组 (5 .2 μg/d)及对照组肺表面的转移结节数分别为 (1 1 9± 70 )个和 (1 4± 1 2 )个 ;对肺转移瘤组织切片免疫组化结果表明给药组转移瘤组织中血管内皮生长因子的表达明显高于对照组 .同时 ,在Mc3细胞和瘤体组织中均发现有雌激素受体的表达 .结论 :生理浓度的雌激素可促进人涎腺黏液表皮样癌血管内皮生长因子的表达
Objective: To study the effect of estrogen on angiogenesis in human salivary gland mucoepidermoid carcinoma.Methods: Immunohistochemistry and nude mice transplantation of lung xenografts were used to investigate the effect of 17β-estradiol on the expression of highly metastatic human mucoepidermoid carcinoma cells The expression of vascular endothelial growth factor (MVD) and the tumorigenic ability of lung transplantation in Mc3 cells.Methods Immunohistochemical method was used to detect the estrogen receptor in Mc3 cells.Results: In vitro, 0.1 μmol / L 17β-estradiol Glycol could promote the expression of vascular endothelial growth factor (VEGF) in Mc3 cells.In the experimental group, the number of metastatic nodules on the lung surface was (112.9 ± 70) in the experimental group (5.2 μg / d) and that in the control group And (14 ± 12), respectively.Immunohistochemical analysis of the tissue sections of lung metastatic tumor showed that the expression of VEGF in the metastatic tumor tissue of the treated group was significantly higher than that of the control group.Meanwhile, in the Mc3 cells and tumor tissues Estrogen receptor expression was found in all cases.Conclusion: Physiological estrogen can promote the expression of vascular endothelial growth factor in human salivary gland mucoepidermoid carcinoma
