目的:采用高效液相色谱法同时测定沙棘果中槲皮素、山柰酚和异鼠李素的含量。方法:Kromasil-C18(250 mm×4.6mm,5μm)色谱柱,流动相为乙腈(A)-0.6%磷酸(B)水溶液,线性梯度洗脱(0~15 min,33%→32%A;15~30 min,32%→31%A),流速1.0mL.min-1,柱温30℃,检测波长370 nm。结果:槲皮素,山柰酚和异鼠李素的质量浓度分别在1.68~26.88,1.52~24.32和1.76~28.16μg.mL-1范围内线性关系良好,平均回收率分别为98.55%,97.63%和97.98%,RSD分别为1.48%,1.66%和1.54%。结论:所建立的方法简单易行,准确可靠,可用于沙棘果中槲皮素、山柰酚和异鼠李素的含量测定。
Objective: To determine the contents of quercetin, kaempferol and isorhamnetin in seabuckthorn by high performance liquid chromatography (HPLC). Methods: Kromasil-C18 (250 mm × 4.6 mm, 5 μm) column was used. The mobile phase consisted of acetonitrile (A) -0.6% 15 ~ 30 min, 32% → 31% A), flow rate 1.0 mL · min-1, column temperature 30 ℃, detection wavelength 370 nm. Results: The calibration curves of quercetin, kaempferol and isorhamnetin were linear in the range of 1.68 ~ 26.88, 1.52 ~ 24.32 and 1.76 ~ 28.16 μg.mL-1, respectively. The average recoveries were 98.55% and 97.63, respectively % And 97.98% respectively, with RSDs of 1.48%, 1.66% and 1.54% respectively. Conclusion: The established method is simple, accurate and reliable, and can be used for the determination of quercetin, kaempferol and isorhamnetin in sea buckthorn fruits.
