目的采用社交应激方式诱导C57BL/6小鼠建立抑郁样模型,并采用多种行为学方法对模型进行评价,以验证模型可靠性。方法将20只雄性C57BL/6(C57)小鼠采用随机数字表法分为正常对照组(正常组,10只)和应激模型组(模型组,10只)。另取CD1(ICR)小鼠(10只)用于社交应激。正常组未予干预措施,模型组每天给予社交应激,连续10天。采用社交接触实验、高架十字实验和悬尾实验对各组小鼠进行行为学评价;采用E LISA法检测小鼠血清皮质醇水平,评估模型可靠性。结果社交接触实验显示,与本组CD1小鼠不存在时比较,正常组小鼠社交区停留时间延长,角落区停留时间缩短(P<0.05),模型组社交区停留时间缩短,角落区停留时间延长(P<0.05);与正常组CD1小鼠存在时比较,模型组社交区停留时间减少,角落区停留时间增多(P<0.05)。与正常组比较,模型组进入开放臂、闭合臂及高架十字总进入次数明显减少(P<0.05),开放臂进入次数比例降低(P<0.05);模型组小鼠在悬尾实验中后4 min内不动时间延长(P<0.05);模型组血清皮质醇水平明显升高(P<0.01)。结论社交应激诱导C57小鼠建立社交应激性抑郁样模型较为可靠,可能适用于中西医结合病证模型的研究。
Objective To establish a model of depression in C57BL / 6 mice by social stress and evaluate the model by various behavioral methods to verify the reliability of the model. Methods Twenty male C57BL / 6 (C57) mice were divided into normal control group (n = 10) and stress model group (n = 10) by random number table. Another CD1 (ICR) mouse (10 mice) was used for social stress. The normal group without intervention, the model group given daily social stress, for 10 days. The social contact experiment, elevated cross test and tail suspension test were used to evaluate the behavior of mice in each group. The serum cortisol level was detected by E-ELISA, and the model reliability was evaluated. Results The social exposure experiments showed that in the normal group, the residence time of the mice in the social area was prolonged and the retention time in the corner area was shortened (P <0.05) compared with that in the absence of the CD1 mice in this group. The residence time in the social area of the model group was shortened, (P <0.05). Compared with the normal control group, the retention time in the social area decreased and the retention time in the corner area increased (P <0.05). Compared with the normal group, the number of open arms in the model group decreased significantly (P <0.05) and the number of open arms decreased (P <0.05). In model group, (P <0.05). The level of serum cortisol in the model group was significantly increased (P <0.01). Conclusion Social-stress-induced C57 mice are more reliable in establishing social stress-induced depression model and may be suitable for the study of integrated TCM-WM disease model.
