目的探讨菟丝子黄酮对氧化损伤的人脐静脉内皮细胞(HUVECs)的保护作用。方法体外培养内皮细胞,采用MTT法测定细胞的增殖,分光光度计法检测一氧化氮(NO)、一氧化氮合酶(NOS)、放射免疫方法测定内皮素-1(ET-1),观察菟丝子黄酮对过氧化氢(H2O2)损伤的HUVECs的影响。结果①菟丝子黄酮(0.1~0.5mg/L)可提高正常培养的HUVECs的细胞活性,且具有浓度依赖性;菟丝子黄酮(0.5mg/L)能明显减轻H2O2对HUVECs所致的氧化损伤,使细胞增殖活性增高((P<0.01)。②H2O2损伤模型组(200mmol/L)与空白组比较,NO释放和NOS活性水平显著下调(P<0.01),ET-1释放水平显著上调(P<0.01),菟丝子(0.5mg/L)+H2O2(200mmol/L)组与H2O2损伤模型组(200 mmol/L)比较,NO释放水平和NOS活性明显上调(P<0.01),ET-1水平显著下调(P<0.01)。结论菟丝子黄酮对H2O2诱导的HUVECs的损伤具有保护作用。
Objective To investigate the protective effects of dodder seeds on oxidative injury of human umbilical vein endothelial cells (HUVECs). Methods The endothelial cells were cultured in vitro. The proliferation of the cells was measured by MTT assay. The levels of nitric oxide (NO), nitric oxide synthase (NOS) and endothelin - 1 (ET - 1) were determined by spectrophotometer. Effect of Dusky Flavin on HUVECs Injured by Hydrogen Peroxide (H2O2). Results ① Dusanum flavonoids (0.1-0.5 mg / L) could increase the cell viability of cultured HUVECs in a concentration-dependent manner. Dusanum flavonoids (0.5 mg / L) could significantly reduce the oxidative damage induced by H2O2 on HUVECs, (P <0.01) .②The level of NO release and NOS activity in H 2 O 2 -induced model group (200 mmol / L) was significantly lower than that in control group (P <0.01), ET-1 release was significantly increased (P <0.01), and the level of ET-1 was significantly down-regulated (P <0.01). Compared with 200 mmol / L H2O2 group, P <0.01) .Conclusion Dodder flavonoids have a protective effect on H2O2-induced HUVECs injury.
