目的探讨榄香烯对体外培养的人胃腺癌细胞株BGC-823中乙酰肝素酶(HPA)表达水平的影响。方法采用免疫细胞化学方法对药物作用后的BGC-823细胞HPA在蛋白水平进行检测。结果榄香烯能明显抑制人胃腺癌细胞株BGC-823的生长并降低HPA表达水平,且有时间剂量依赖性。结论榄香烯通过降低胃腺癌细胞HPA蛋白表达水平,从而遏制了肿瘤血管、淋巴管转移,为肿瘤靶向治疗提供了理论基础和实验依据。
Objective To investigate the effect of elemene on the expression of heparanase (HPA) in human gastric adenocarcinoma cell line BGC-823 cultured in vitro. Methods Immunocytochemistry was used to detect the protein level of HPA in BGC-823 cells after drug treatment. Results Elemene significantly inhibited the growth of human gastric adenocarcinoma cell line BGC-823 and decreased the expression of HPA, which was time-and dose-dependent. Conclusion Elemene can inhibit the tumor blood vessel and lymphatic metastasis by reducing the expression of HPA protein in gastric adenocarcinoma cells and provide the theoretical and experimental basis for tumor targeted therapy.
