目的 通过用 Wistar大鼠制造眼钝挫伤模型 ,观察虹膜毛细血管通透性变化 ,探讨其与外伤性前房出血后再次出血的关系。方法 选用 5 0只 Wistar大鼠麻醉后用钝器敲击其双眼造成前房出血的眼外伤模型 ,每天观察出血情况且在损伤后 1、3、5、7、9、11d分别取 2只眼虹膜组织制备石蜡切片 (HE染色 )作光镜检查 ,另外在按 2 m L· kg- 1 剂量将10 g· L- 1 Evan蓝溶液注入大鼠的左心室然后经左心室心脏灌洗后取出虹膜组织按一定剂量浸泡于甲酰胺溶液中 ,于 45℃恒温水浴 72 h,取上清液 ,于 6 2 0 nm波长分光光度计测定比色值标记成一曲线 ;并测不同浓度 (0 .0 1~ 0 .1g· L- 1 ) Evan蓝溶液中在 6 2 0 nm波长的透光度 ,制作一标准曲线。结果 外伤后 ,虹膜毛细血管通透性均增高 ,增高程度在不同时间不相同。在 19只再次出血的标本中 ,其虹膜通透性均保持高水平。结论 再次出血大鼠中 ,发现所测得的透光度维持在高水平 ,于组织间隙仍可见血细胞游离其间 ,说明毛细血管壁的不稳定状态是造成再次出血的原因。
OBJECTIVE: To observe the changes of iris capillaries permeability by using the Wistar rat model of blunt traumatic eye injury and to explore its relationship with rebleeding after traumatic hyphema. Methods Fifty Wistar rats were anesthetized with a blunt instrument to knock on their eyes to cause the model of ocular trauma in the anterior chamber hemorrhage. The hemorrhage was observed daily and two eyes were taken at 1, 3, 5, 7, 9 and 11 days after injury The iris tissue was made into paraffin sections (HE staining) for light microscopy. In addition, 10 g · L -1 Evan blue solution was injected into the left ventricle of rats at a dose of 2 m L · kg -1 and then left ventricular cardioplegia Iris tissue soaked in formamide solution according to a certain dose, at 45 ℃ water bath for 72 h, the supernatant, measured at 620 nm wavelength spectrophotometer colorimetric value marked a curve; and measured at different concentrations (0 .0 1 ~ 0. 1g · L- 1) Evan blue solution at a wavelength of 6 2 0 nm transmittance, making a standard curve. Results After trauma, iris capillaries permeability increased, the degree of increase in different time is not the same. In the 19 rebleeding specimens, iris permeability remained high. Conclusion In the rebleeding rat, the measured transmittance was found to be maintained at a high level. Blood cells could still be seen in interstitial spaces, indicating that the capillary wall instability was the cause of rebleeding.
