目的:探索体内、体外应用不同干预因素对系统性红斑狼疮(SLE)患者自然杀伤T细胞(NKT)的含量变化的影响,为SLE患者治疗寻求一条新的途径。方法:采用流式细胞术(FCM)检测30例SLE患者体内外应用不同干预因素前后外周血Vα24+Vβ11+、CD161+Vα24+NKT细胞含量。结果:SLE患者体内应用甲强龙及甲强龙加丙球24h后Vα24+Vβ11+、CD161+Vα24+NKT细胞占淋巴细胞比率明显高于用药前(P<0.01),SLE患者体外制备PBMC后加甲强龙、甲强龙加丙球及IL-12+IL-18进行细胞培养48h后,Vα24+Vβ11+、CD161+Vα24+NKT细胞占淋巴细胞比率明显高于用药前(P<0.01)。结论:体内外应用甲强龙及甲强龙加丙球后,Vα24+Vβ11+、CD161+Vα24+NKT细胞占淋巴细胞比率明显高于用药前;体外加IL-12+IL-18后与用药前比,Vα24+Vβ11+、CD161+Vα24+NKT细胞占淋巴细胞比率明显高于用药前;表明细胞因子IL-12+IL-18可能成为治疗SLE的一种新的方法。
Objective: To explore the effects of different interventions in vitro and in vivo on the content of natural killer T cells (NKT) in patients with systemic lupus erythematosus (SLE) and to find a new way for the treatment of SLE patients. Methods: The levels of Vα24 + Vβ11 + and CD161 + Vα24 + NKT in peripheral blood of 30 patients with SLE before and after different interventions were detected by flow cytometry (FCM). Results: The ratio of Vα24 + Vβ11 +, CD161 + Vα24 + NKT cells to lymphocytes in patients with SLE was significantly higher than that before medication (P <0.01), and PBMCs were prepared from SLE patients in vitro The ratio of Vα24 + Vβ11 +, CD161 + Vα24 + NKT cells to lymphocytes was significantly higher than that before treatment (P <0.01) after 48h treatment with methylprednisolone, methylprednisolone plus corticosteroid and IL-12 + IL-18. Conclusion: The ratio of Vα24 + Vβ11 +, CD161 + Vα24 + NKT cells to lymphocytes was significantly higher than that before treatment with methylprednisolone plus methylprednisolone plus propofol in vitro and in vivo. Before addition of IL-12 + IL- The ratios of Vα24 + Vβ11 +, CD161 + Vα24 + NKT cells to lymphocytes were significantly higher than those before treatment. It is suggested that cytokines IL-12 + IL-18 may be a new method for the treatment of SLE.
