目的:初步探讨CD4+CD25+/highCD127lowT调节性T细胞(Tregs)及TGF-β在不同病程阶段多发性骨髓瘤(MM)患者外周血中的表达水平,并研究患者血清TGF-β水平是否与CD4+CD25+/highCD127lowTregs具有相关性。方法:流式细胞术(FCM)检测50例MM患者及30例健康志愿者外周静脉血CD4+CD25+/highCD127lowTregs细胞的比例并分析比较。ELISA法定量检测血清TGF-β的表达水平。结果:(1)化疗后组CD4+CD25+CD127low、CD4+CD25highCD127lowT细胞的比例均显高于初诊组及正常对照组;初诊组与正常对照组的CD4+CD25+CD127lowT细胞相比无统计学意义,但其CD4+CD25highCD127lowT细胞明显高于正常对照组。(2)化疗后稳定期CD4+CD25+CD127lowT细胞与活动期没有差异,但其CD4+CD25highCD127lowT细胞明显低于活动期。(3)MM各组TGF-β水平均较对照组明显升高,但各组比较无统计学意义。(4)TGF-β与CD4+CD25+CD127low、CD4+CD25highCD127lowT细胞的表达水平均不成相关性。结论:CD4+CD25+/highCD127low调节性T细胞MM患者外周血中比例明显升高,且化疗及疾病进展程度可能影响其在外周血的比例。TGF-β可能不是导致其比例增加的直接原因。
Objective: To investigate the expression of CD4 + CD25 + / highCD127lowT regulatory T cells (Tregs) and TGF-β in peripheral blood of patients with multiple myeloma (MM) at different stages of disease and to investigate whether serum TGF- + CD25 + / highCD127lowTregs. Methods: The proportion of CD4 + CD25 + / highCD127low Tregs in peripheral blood of 50 MM patients and 30 healthy volunteers was detected by flow cytometry (FCM) and analyzed. ELISA quantitative detection of serum TGF-β expression levels. Results: (1) The proportion of CD4 + CD25 + CD127low and CD4 + CD25highCD127low T cells in the chemotherapy group were significantly higher than those in the newly diagnosed group and the normal control group. There was no significant difference in the CD4 + CD25 + CD127lowT cells between the newly diagnosed group and the normal control group , But its CD4 + CD25highCD127lowT cells were significantly higher than the normal control group. (2) The stable CD4 + CD25 + CD127lowT cells after chemotherapy did not differ from the active phase, but the CD4 + CD25highCD127lowT cells were significantly lower than the active phase. (3) The levels of TGF-β in MM group were significantly higher than those in control group, but there was no significant difference among the three groups. (4) There was no correlation between TGF-β and CD4 + CD25 + CD127low, CD4 + CD25highCD127lowT cells. Conclusion: The proportion of CD4 + CD25 + / highCD127low regulatory T cells in peripheral blood is significantly increased, and the degree of chemotherapy and disease progression may affect the proportion of peripheral blood. TGF-β may not be the direct cause of its increased proportion.
