目的:研究半枝莲提取物对肺癌A549细胞生存素(survivin)蛋白表达的影响及其作用机制。方法:体外培养(10%胎牛血清、1640培养基)肺癌A549细胞,MTT法观察半枝莲提取物对A549细胞的抑制作用,TUNEL法检测细胞凋亡,免疫细胞化学(SP)法检测半枝莲提取物对A549细胞survivin蛋白表达的影响。结果:MTT法结果显示,半枝莲提取物质量浓度5~40 mg·L-1对A549细胞的抑制率呈浓度依赖性(P<0.01)。半枝莲提取物18.86 mg·L-1剂量组凋亡指数为(38.40±7.41)%,对照组为(6.22±0.45)%,两者有显著性差异(P<0.01)。半枝莲提取物组survivin蛋白阳性细胞数明显低于对照组(P<0.01)。结论:半枝莲提取物能明显抑制肺癌A549细胞生长,其机制可能与下调survivin表达有关。
Objective: To study the effect of Scutellaria barbata extract on the expression of survivin protein in lung cancer A549 cells and its mechanism. Methods: Human lung adenocarcinoma A549 cells were cultured in vitro (10% fetal bovine serum and 1640 medium). The inhibitory effects of Scutellaria barbata extract on A549 cells were observed by MTT assay. The apoptosis of A549 cells was detected by TUNEL assay. Effect of extract of Scutellaria barbata on survivin protein expression in A549 cells. Results: MTT assay showed that the inhibitory rate of Scutellaria barbata extract concentration of 5 ~ 40 mg · L-1 on A549 cells in a concentration-dependent manner (P <0.01). The apoptosis index of Scutellaria barbata extract (18.86 mg · L -1) was (38.40 ± 7.41)% and that of control group was (6.22 ± 0.45)%, both of which were significantly different (P <0.01). Scutellaria barbata extract group survivin protein positive cells was significantly lower than the control group (P <0.01). Conclusion: Scutellaria barbata extract can significantly inhibit the growth of lung cancer A549 cells, the mechanism may be related to down-regulation of survivin expression.
