目的:探讨血卟啉单甲醚(HMME)光动力治疗卵巢癌的体外治疗效果。方法:不同浓度(0~20μg/ml)HMME联合不同能量激光(0~6J/cm2)处理大鼠上皮性卵巢癌细胞株NuTu-19细胞后,MTT比色法检测细胞存活率;Annexin V/PI双染色,流式细胞仪分析细胞死亡方式。结果:HMME无暗毒性,随着药物浓度与激光剂量的增加,细胞存活率逐渐下降,大剂量可取得完全杀伤的效果,细胞死亡方式以坏死为主。结论:HMME对卵巢癌细胞具有明确的体外光动力学杀伤效应。
Objective: To investigate the in vitro therapeutic effect of hematoporphyrin monomethyl ether (HMME) photodynamic therapy on ovarian cancer. METHODS: NuTu-19 cells were treated with HMME combined with different energy laser (0 ~ 6J / cm2) at different concentrations (0 ~ 20μg / ml) for MTT assay. Annexin V / PI double staining, flow cytometry analysis of cell death patterns. Results: HMME showed no toxicity. With the increase of drug concentration and laser dose, the cell survival rate decreased gradually and the complete killing effect could be obtained at high dose. The necrosis of cells was the main way. CONCLUSION: HMME has a clear in vitro photodynamic killing effect on ovarian cancer cells.
