目的:研究中药温化蠲痹方对胶原诱导性关节炎(CIA)大鼠滑膜白细胞介素-1β(IL-1β)、膜白细胞介素-8(IL–8)表达的影响,探讨温化蠲痹方治疗CIA的作用机制。方法:选用健康雄性Wistar大鼠,随机分为造模组、正常对照组(10只,简称正常组),造模组采用牛Ⅱ型胶原乳剂于大鼠尾根部注射,建立CIA模型。再选取造模成功大鼠30只随机分为模型组(10只)、MTX组(10只)、温化蠲痹方组(10只,简称中药组)。中药组给予温化蠲痹方22.9 g/kg·d灌胃,模型组灌胃生理盐水,均每天1次,MTX组按0.78 mg/kg剂量灌胃MTX混悬液,每周1次,连续30 d。给药结束后,运用免疫组化法检测大鼠滑膜IL-1β、IL–8表达水平。结果:与模型组比较,治疗后中药组和MTX组大鼠关节炎指数(AI)评分明显降低(P<0.01);与治疗前比较,中药组和MTX组大鼠AI评分均明显降低(P<0.01)。与正常组比较,模型组大鼠滑膜IL-1β、IL–8表达水平明显升高(P<0.01);与模型组比较,中药组和MTX组大鼠滑膜IL-1β、IL–8表达水平明显降低(P<0.01),中药组和MTX组之间比较差异无统计学意义(P>0.05)。结论:中药温化蠲痹方可能通过下调CIA大鼠滑膜IL-1β、IL–8水平达到而治疗CIA目的。
Objective: To study the effect of Wenhua Wanbi Recipe on the expression of interleukin-1β (IL-1β) and membrane interleukin-8 (IL-8) in synovium of rats with collagen-induced arthritis (CIA) Huayu Bi Fang treatment of CIA mechanism of action. Methods: Healthy male Wistar rats were randomly divided into model group and normal control group (n = 10, referred to as normal group). The model rats were injected with the tail of the rats with type Ⅱ collagen emulsion to establish CIA model. Thirty rats were selected and randomly divided into model group (n = 10), MTX group (n = 10) and Wenhuahubi group (n = 10). The rats in the traditional Chinese medicine group were treated with 22.5 g / kg · d of Wenhua Xiebi Decoction, and the model group was given normal saline once a day. The MTX group was orally administered with MTX suspension at a dose of 0.78 mg once per week for continuous 30 d. After the administration, the expression of IL-1β and IL-8 in synovium of rats was detected by immunohistochemistry. Results: Compared with the model group, the arthritis index (AI) scores of the Chinese medicine group and the MTX group were significantly decreased after treatment (P <0.01). Compared with the pretreatment group, the AI scores of the Chinese medicine group and the MTX group were significantly decreased <0.01). Compared with the normal group, the expression of IL-1β and IL-8 in the synovium of the model group was significantly increased (P <0.01). Compared with the model group, IL-1β, IL-8 (P <0.01). There was no significant difference between MTX group and MTX group (P> 0.05). Conclusion: The traditional Chinese medicine WENHUA PUBEI may reduce the CIA rats synovium IL-1β, IL-8 levels to achieve the purpose of CIA.
