取中国四川产的大黄(雅黄)710g,50%含水丙酮室温提取,乙酸乙酯-水萃取,水层酸化,放置去深棕色沉淀.取上清提取物经浓缩后进行Sephadex LH-20柱层析,水洗脱,得总苷,甲醇溶解,溶解部分再经ODS-Q3柱层析,用含醋酸(10ml/L)的5~15%乙腈溶液洗脱,得大黄酸苷(Rheinosides)A(14.18g),B(3.64g),C(2.64g)和D(4.56g).经NMR分析和化学降解,鉴定A和B为一对差向异构体,结构为:8-O-β-D-萄萄糖基-10-羟基-10-C-β-D-葡萄糖基大黄-9-蒽酮;C和D为另一对差向异构体,结构为:8-O-β-D-葡萄糖基-10-C-β-D-葡萄糖基大黄9-蒽酮,它们的绝对构型正在鉴定中.
