目的利用25对AFLP引物组合对DDK品系小鼠与实验室常用的3个小鼠品系进行研究,分析DDK品系小鼠的遗传种质性,为解决其独特繁殖现象与遗传背景的关系做好理论准备。方法根据AFLP分析结果,统计每个引物组合在各个品系中的多态性条带数和特异性条带数,计算DDK小鼠与其他实验室常用小鼠(C57BL/6J、BALB/c、PWK)之间的遗传相似系数和遗传距离。结果 25对AFLP引物组合共检测到1 224个条带,多态性条带有279条,占总条带的22.79%。结论 DDK品系小鼠与BALB/c小鼠品系遗传关系最近,与C57BL/6J品系关系较近;DDK品系小鼠与PWK品系小鼠遗传关系最远;本研究为进一步探讨DDK小鼠的独特繁殖现象提供了理论依据;同时也阐明了AFLP技术在物种鉴定、遗传背景分析及预测亲本杂交效果等方面具有明显的优越性。
OBJECTIVE: To study the genetic germplasm of DDK strain mice using 25 pairs of AFLP primer combinations and to study the relationship between the unique breeding phenomenon and genetic background ready. Methods Based on the results of AFLP analysis, the number of polymorphic bands and the number of specific bands of each primer combination in each strain were calculated and compared with other commonly used laboratory mice (C57BL / 6J, BALB / c, PWK ) Genetic similarity coefficient and genetic distance between. Results A total of 1 224 bands were detected in 25 pairs of AFLP primer combinations. There were 279 polymorphic bands, accounting for 22.79% of the total bands. Conclusion DDK strain has the closest genetic relationship with BALB / c mouse strain, and has a closer relationship with C57BL / 6J strain. DDK strain mouse has the furthest genetic relationship with PWK strain. In order to further explore the unique breeding of DDK mouse, Phenomenon provides a theoretical basis; also clarified the AFLP technology in species identification, genetic background analysis and prediction of the effect of parental hybrid has obvious advantages.
