目的:探讨高脂饮食对大鼠肾组织中炎症因子抑瘤素-M(OSM)表达的影响。方法:将健康雄性Wistar大鼠随机分为普通饮食组、高脂饮食4周组、高脂饮食8周组、高脂饮食12周组4组,应用蛋白免疫印迹技术检测各组大鼠肾组织中OSM的表达变化,ELISA法检测各组大鼠血清中OSM表达,生化方法检测肾功能、血脂及24 h尿蛋白定量,对各指标与OSM表达进行相关性分析。结果:高脂血症大鼠血清中OSM表达较对照组明显增高(P<0.05),且随着高脂饮食时间的延长,OSM表达亦明显升高。生化检测提示,随高脂饮食时间的增加,肾功能、血脂、24 h尿蛋白定量逐渐增加,OSM表达与肾功能、血脂、24 h尿蛋白定量呈显著正相关(P<0.05)。结论:高脂环境下大鼠肾组织OSM表达明显增加,且与肾功能、血脂、24 h尿蛋白定量明显相关,提示OSM表达很可能参与高脂致肾损害的病理过程。
Objective: To investigate the effect of high-fat diet on the expression of inflammatory cytokine-M (OSM) in rat kidney. Methods: Healthy male Wistar rats were randomly divided into normal diet group, high-fat diet for 4 weeks, high-fat diet for 8 weeks and high-fat diet for 12 weeks for 4 groups. Western blotting was used to detect the changes of renal tissue The OSM expression was detected by ELISA. The serum levels of OSM in each group were detected by ELISA. The renal function, blood lipid and 24 h urinary protein were detected by biochemical methods. The correlation between OSM expression and each index was analyzed. Results: The serum OSM expression in hyperlipidemic rats was significantly higher than that in the control group (P <0.05), and the expression of OSM was also increased with the prolongation of high fat diet. The biochemical tests showed that with the increase of the time of high-fat diet, the renal function, blood lipid and 24 h urinary protein increased gradually. The expression of OSM was positively correlated with renal function, blood lipid and 24 h urine protein (P <0.05). CONCLUSIONS: The expression of OSM in rat kidney increased significantly under high-fat environment and significantly correlated with renal function, blood lipid and 24-h urinary protein, suggesting that OSM expression may be involved in the pathogenesis of hyperlipemia-induced renal damage.
