目的:筛选用来富集齿叶白鹃梅叶总黄酮的树脂型号,并确定最佳纯化工艺。方法:通过考察各种树脂对总黄酮的吸附和解吸附情况确定最佳树脂型号,对上样浓度、洗脱溶剂等因素的考察,确定最佳工艺参数。结果:最适宜树脂为D101型大孔树脂,上样溶液中总黄酮的浓度以2.5mg.mL-1为宜,最大上样量为1.5倍树脂床体积,洗脱溶剂为90%乙醇,洗脱剂用量为4.5倍树脂床体积。结论:采用此工艺纯化后的总黄酮的含量可达到59.1%。
OBJECTIVE: To screen the resin type used to enrich the total flavonoids of Rhododendron fortunei leaves and determine the best purification process. Methods: The optimum resin type was determined by examining the adsorption and desorption of total flavonoids by various resins. The optimal parameters were determined by the investigation of factors such as the concentration of sample and the elution solvent. Results: The optimum resin was D101 macroporous resin. The concentration of total flavonoids in the sample solution was 2.5mg.mL-1, the maximum sample volume was 1.5 times the volume of resin bed, the elution solvent was 90% ethanol, washed The amount of release agent is 4.5 times the volume of the resin bed. Conclusion: The content of total flavonoids purified by this process can reach 59.1%.
