目的:利用D101型大孔吸附树脂建立分离纯化藜麦总皂苷的工艺参数。方法:采用薄层色谱法确定D101大孔吸附树脂对藜麦总皂苷的动态吸附容量和洗脱条件,采用紫外可见分光光度法测定藜麦总皂苷含量,确定最佳的吸附洗脱工艺。结果:D101型大孔吸附树脂对藜麦总皂苷有较好的吸附分离性能,经过大孔树脂纯化处理,藜麦药液干浸膏中藜麦总皂苷含量由24.9%提高到53.9%,干浸膏得率由47.3%下降至15.0%。如果在60%乙醇洗脱前,增加20%乙醇洗脱操作以除去部分非皂苷部分,藜麦总皂苷含量可进一步提高到72.3%。结论:D101型大孔吸附树脂对藜麦总皂苷纯化效果较好。“,”Objective:To establish the process parameters for the separation and purification of total saponins from quinoa by D101 macroporous adsorption resin. Methods:the dynamic adsorption capacity and elution conditions of D101 macroporous adsorption resin for quinoa saponins were determined by thin layer chromatography. The content of quinoa saponins was determined by UV-vis spectrophotometry to determine the best adsorption and elution process. Results:D101 macroporous resin had good adsorption and separation performance for quinoa total saponins. After purification with macroporous resin, the content of quinoa total saponins in quinoa liquid dry extract increased from 24.9% to 53.9%, and the yield of dry extract decreased from 47.3% to 15.0%.If 20% ethanol elution operation is added to remove some non saponin parts before 60% ethanol elution, the total saponin content of quinoa can be further increased to 72.3%. Conclusion:D101 macroporous adsorption resin has a good effect on the purification of total saponins from quinoa.
