目的 探讨松果体功能减退致褪黑素 (MT)分泌减少对大鼠学习记忆能力及海马星形胶质细胞的影响。方法 将大鼠进行 Y型迷宫测试 ,淘汰学习障碍的大鼠 ,将学习正常的大鼠随机分三组 ,实验组手术摘除松果体 ,对照组给予假手术 ,空白对照组不处理 ,饲养 40天后再行 Y型迷宫测试 ,以胶质纤维酸蛋白 (GFAP)标记星形胶质细胞 ,SP法检测。结果 实验组大鼠学习能力明显低于对照组大鼠 (P <0 .0 1) ,海马 GFAP免疫反应阳性细胞减少 ,分布稀疏 ,与对照组和空白组相比均 P <0 .0 1。结论 大鼠松果体摘除可以引起大鼠学习记忆障碍 ,其原因可能与海马结构星形胶质细胞减少 ,神经元微环境调节能力减退 ,海马结构易损伤有关
Objective To investigate the effect of decreased secretion of melatonin (MT) on learning and memory ability and hippocampal astrocytes in rats with dysfunction of pineal gland. Methods The rats were subjected to Y-maze test and the rats with learning disability were eliminated. The normal rats were divided into three groups at random. The pineal glands were excised in the experimental group and the sham operation in the control group. The rats in the blank control group were not treated. Day after the Y-maze test, with glial fibrillary acidic protein (GFAP) labeled astrocytes, SP test. Results The learning ability of the experimental group was significantly lower than that of the control group (P <0.01). The GFAP immunoreactive positive cells in the hippocampus decreased and were sparsely distributed, compared with the control group and the blank group (P <0.01). Conclusions The rat pineal gland ablation can cause learning and memory impairment in rats. The reasons may be related to the decrease of hippocampal astrocytes, the diminished ability of neuron microenvironmental regulation and the easy damage of hippocampal formation
