目的:探讨应用丹芍化纤胶囊加食苹果对大鼠肝纤维化tsp-1、TGF-β1的影响。方法:雄性健康SD大鼠46只,采用四氯化碳(CCl4)、高脂饮食加乙醇混合因素造模成功,随机分3组,分别给予相应药物及苹果汁灌胃观察8周。实验结束后RT-PCR检测血小板反应蛋白(thrombospondin-1,tsp-1)mRNA的表达,免疫组织化学技术检测肝组织转化生长因子β1(transforming growth factor,TGF-β1)的表达。结果:RT-PCR检测在正常对照组中tsp-1 mRNA有极少量表达,与正常对照组比较,肝纤维化模型组、自然恢复组tsp-1 mRNA表达显著升高(P<0.01),治疗8周后,丹芍组tsp-1 mRNA表达下降(P<0.05),丹苹组下降更显著(P<0.01)。免疫组化结果示正常肝脏组织只见很少量TGF-β1表达,治疗8周后与自然恢复组比较,丹芍组TGF-β1表达明显下降(P<0.05),丹苹组与下降更显著(P<0.01)。结论:应用丹芍化纤胶囊加食苹果能明显改善大鼠肝功能,降低肝组织tsp-1 mRNA和TGF-β1的表达,具有明显的辅助抗肝纤维化作用。
Objective: To investigate the effect of DanShaoHuaXian capsule and apple on tsp-1 and TGF-β1 of liver fibrosis in rats. Methods: Forty-six male Sprague-Dawley rats were randomly divided into three groups. The rats were given GCl4, high-fat diet and ethanol for 8 weeks respectively. The mRNA expression of thrombospondin-1 (tsp-1) was detected by RT-PCR and the expression of transforming growth factor-β1 (TGF-β1) was detected by immunohistochemistry. Results: Compared with the normal control group, the expression of tsp-1 mRNA in the normal control group was significantly increased (P <0.01) by RT-PCR, and the expression of tsp-1 mRNA in the liver fibrosis model group and the natural recovery group was significantly increased After 8 weeks, the expression of tsp-1 mRNA in Danshen group was decreased (P <0.05), and decreased in Danshen group (P <0.01). Immunohistochemical results showed that only a small amount of TGF-β1 expression was observed in normal liver tissue. After 8 weeks of treatment, compared with the natural recovery group, the expression of TGF-β1 in Dan Shao Decoction group decreased significantly (P <0.05) (P <0.01). Conclusion: Application of Danshaohuaxian Capsule to the apple can obviously improve the liver function of rats and reduce the expression of tsp-1 mRNA and TGF-β1 in liver tissue, and has obvious anti-hepatic fibrosis effect.
