目的:观察川崎病血清对血管内皮细胞表达血小板源生长因子B链蛋白(PDGF-B)的影响,以及川崎病血清作用后的内皮细胞条件培养基对血管平滑肌细胞表达PDGF受体信使核糖核酸(mRNA)和对细胞周期的影响。方法:采用免疫细胞化学、核酸杂交和流式细胞分析方法。结果:川崎病血清能够强烈诱导内皮细胞表达PDGF-B;川崎病血清作用后的内皮细胞条件培养基能够显著促进平滑肌细胞表达PDGF受体mRNA,并强烈诱导平滑肌细胞增殖。结论:PDGF—PDGF受体途径的激活可能参与了川崎病冠状动脉并发症的形成。
Objective: To observe the effect of Kawasaki disease serum on the expression of platelet-derived growth factor B-chain protein (PDGF-B) in vascular endothelial cells and the conditioned medium of endothelial cells after Kawasaki disease serum on the expression of PDGF receptor messenger ribonucleic acid mRNA) and its effect on cell cycle. Methods: Immunocytochemistry, nucleic acid hybridization and flow cytometry were used. Results: Kawasaki disease serum strongly induced PDGF-B expression in endothelial cells; Kawasaki disease serum-conditioned endothelial cells conditioned medium significantly promoted PDGF receptor mRNA expression in smooth muscle cells and strongly induced proliferation of smooth muscle cells. Conclusion: The activation of PDGF-PDGF receptor pathway may be involved in the formation of coronary artery disease in Kawasaki disease.
