目的观察缺血/缺氧(I-H)预处理在子宫内膜异位症中的作用及机制。方法诊断子宫肌瘤需行子宫切除手术患者20例,月经周期正常,病理证实子宫内膜处于分泌早期。术中分离双侧子宫动脉,采用无损伤血管夹钳夹/开放子宫动脉各5min,重复3次。I-H预处理前(对照组)、后(实验组)刮取前壁子宫内膜,种植于鸡胚尿囊膜(CAM),72h后观察种植内膜成活率。免疫组化检测内膜组织中基质金属蛋白酶1(MMP-1)的表达。结果成功构建了鸡胚模型。实验组种植灶中MMP-1的含量高于对照组(P<0.05)。结论 I-H预处理上调子宫内膜MMP-1的表达,提高CAM模型中种植成活率。
Objective To observe the effect and mechanism of ischemic / hypoxic (I-H) preconditioning in endometriosis. Methods Diagnosis of uterine fibroids need hysterectomy in patients with 20 cases of normal menstrual cycle, endometrial pathology confirmed early secretion. Intraoperative separation of bilateral uterine artery, undamaged vascular clamp / open uterine artery 5min, repeated 3 times. Before I-H preconditioning (control group), the anterior wall endometrium was scraped from the anterior wall of the chick embryo and implanted into the chorioallantoic membrane (CAM) of the chick embryo. After 72 hours, the survival rate of the intima was observed. Immunohistochemistry was used to detect the expression of matrix metalloproteinase-1 (MMP-1) in the endometrium. Results The chicken embryo model was constructed successfully. The content of MMP-1 in experimental group was higher than that in control group (P <0.05). Conclusion I-H pretreatment up-regulates the expression of MMP-1 in the endometrium and improves the survival rate of CAM in the model.
