目的 探讨鼻咽癌 (NPC)中p5 3蛋白表达情况及其与临床生物学行为、p5 3基因突变、EB病毒感染的的关系。方法 采用免疫组化检测p5 3蛋白 ,聚合酶链反应 -单链构像多态性 (PCR -SSCP)技术检测p5 3突变 ,聚合酶链反应 (PCR)检测EBV。结果 43例NPC中p5 3蛋白总的阳性表达率为 76 7%( 3 3 / 43 ) ,临床Ⅲ、Ⅳ期中p5 3阳性率为 80 6%( 2 5 / 3 1)明显高于临床Ⅰ、Ⅱ期66 6%( 8/ 12 ) (P <0 0 5 ) ,且表达强度随肿瘤分期而增加 ;NPC伴颈淋巴结转移患者p5 3表达阳性率 88 9%( 2 2 / 2 6) ,明显高于不伴颈淋巴结转移患者p5 3表达阳性率 5 3 5 %( 11/ 17) (P <0 0 1) ;NPC组织中p5 3基因突变率为 4 7%( 2 / 43 ) ,鼻咽炎症组织中无突变 ;EBV阳性率为 93 %( 4 0 / 43 )。结论 NPC组织中p5 3蛋白表达与p5 3基因突变无关 ,与EB病毒的感染有关。p5 3阳性易发生颈淋巴结转移 ,随着NPC病程的发展p5 3的阳性表达率及表达强度均增加
Objective To investigate the expression of p5 3 protein in nasopharyngeal carcinoma (NPC) and its relationship with clinical biological behavior, p5 3 gene mutation and Epstein-Barr virus (EBV) infection. Methods The p5 3 protein was detected by immunohistochemistry and the p5 3 mutation was detected by polymerase chain reaction and single strand conformation polymorphism (PCR-SSCP). The EBV was detected by polymerase chain reaction (PCR). Results The positive rate of p5 3 protein in 43 cases of NPC was 76.7% (3 3/43), and the positive rate of p5 3 in stage Ⅲ and Ⅳ was 80 6% (2 5/3 1) The positive expression rate of p53 in NSCLC patients with cervical lymph node metastasis was 88.9% (22/26), which was significantly higher than that in patients with cervical lymph node metastasis (66.6%, 8/12) (P <0.05) The positive rate of p53 expression was 53.5% (11/17) in patients without lymph node metastasis (P <0.01). The mutation rate of p5 3 gene in NPC was 47% (2/43) There was no mutation in the disease tissues; the positive rate of EBV was 93% (40/43). Conclusion The expression of p5 3 in NPC is not related to the mutation of p5 3 gene, which is related to the infection of Epstein-Barr virus. p5 3 positive cervical lymph node metastasis easily, with the development of NPC course of p5 3 positive expression rate and intensity were increased
