目的 :探讨结直肠癌中长链基因间非编码RNAs(long intergenic non-coding RNAs,lincRNAs)的表达特征。方法 :采用基因芯片技术检测4例结直肠癌及癌旁正常黏膜组织中长链非编码RNAs(long non-coding RNAs,lncRNAs)与mRNAs的表达情况,筛选出差异表达的lincRNAs与mRNAs并用MeV 4.6软件行聚类分析;进一步采用上海伯豪生物技术有限公司的生物芯片分析系统(SBC Analysis System,SAS)分析其相关信号转导通路;采用实时荧光定量-PCR方法验证linc-UCC、linc-DCC与MIR143HG在癌组织及癌旁组织中的表达情况,并分析与结直肠癌临床特征之间的相关性。结果 :采用基因芯片共检测到42 545个RNA,其中124个lincRNAs和1 583个mRNAs的表达有明显差异。生物信息学分析结果显示,有29条信号转导通路可能与这些lincRNAs有关。linc-UCC在较多结直肠癌组织中表达增多,其高表达与肿瘤淋巴结转移密切相关;而linc-DCC与MIR143HG在多数结直肠癌组织中表达减少,二者呈正相关性。结论 :通过基因芯片技术筛选获得与结直肠癌相关的lincRNAs,以期为结直肠癌的临床诊断及治疗提供一些新的靶点。
Objective: To investigate the expression characteristics of long-term intergenic non-coding RNAs (lincRNAs) in colorectal cancer. Methods: The expression of long-chain non-coding RNAs (mRNAs) and mRNAs in 4 colorectal cancer tissues and adjacent normal mucosa tissues were detected by gene chip technique. The differentially expressed lincRNAs and mRNAs were screened out and compared with MeV 4.6 Software cluster analysis. The SBC analysis system (SAS) was further used to analyze the signal transduction pathways. The real-time fluorescence quantitative-PCR method was used to validate linc-UCC, linc-DCC And MIR143HG in cancer tissues and paracancerous tissues, and to analyze the correlation with clinical features of colorectal cancer. Results: A total of 42 545 RNAs were detected by gene chip, of which 124 lincRNAs and 1 583 mRNAs were significantly different. Bioinformatics analysis showed that there are 29 signal transduction pathways may be related to these lincRNAs. The expression of linc-UCC increased in more colorectal cancer tissues, and the high expression of linc-UCC was closely related to tumor lymph node metastasis. However, the expression of linc-DCC and MIR143HG decreased in most colorectal cancer tissues. Conclusion: The lincRNAs related to colorectal cancer were screened by gene chip technology in order to provide some new targets for the clinical diagnosis and treatment of colorectal cancer.
