目的探讨人类脐带间充质干细胞(hUC-MSCs)对肝内胆管癌细胞生长的影响,深入了解其分子机制。方法采用hUC-MSCs条件培养基作用于肝内胆管癌HCCC-9810细胞,通过Transwell共培养实验、MTT比色实验、DNA断裂实验等方法检测细胞增殖和细胞凋亡,通过Western blotting及免疫荧光染色方法检测相关凋亡通路中蛋白质表达水平。结果间充质干细胞条件培养基对HCCC-9810细胞的增殖抑制表现出时间和剂量依赖性,且抑制作用是通过诱导细胞凋亡介导,增殖抑制率由12.87%升至50.98%,凋亡率由10.1%升至49.67%。p-Akt、p-GSK-3β表达下调。结论人类脐带间充质干细胞条件培养基可抑制肝内胆管癌细胞的生长,其分子机制与抑制PI3K/Akt信号通路有关。
Objective To investigate the effect of human umbilical cord mesenchymal stem cells (hUC-MSCs) on the growth of intrahepatic cholangiocarcinoma cells and to understand its molecular mechanism. Methods The hUC-MSCs conditioned medium was used to effect the intrahepatic cholangiocarcinoma HCCC-9810 cells. The cell proliferation and apoptosis were detected by Transwell co-culture assay, MTT colorimetric assay and DNA fragmentation assay. Western blotting and immunofluorescence staining Methods The expression of protein in apoptosis pathway was detected. Results The proliferation of HCCC-9810 cells induced by MSCs conditioned medium was time-and dose-dependent, and the inhibition was mediated by induction of apoptosis. The proliferation inhibition rate increased from 12.87% to 50.98%, and the apoptosis rate Rose from 10.1% to 49.67%. p-Akt, p-GSK-3βexpression downregulation. Conclusion Human umbilical cord mesenchymal stem cell conditioned medium can inhibit the growth of intrahepatic cholangiocarcinoma cells. The molecular mechanism is related to the inhibition of PI3K / Akt signaling pathway.
