作者从1例双轻链病患者尿中用硫酸铵盐析提取的本周氏蛋白(BJP),其K和λ链双扩滴度均高于1:640,而五类Ig阴性。琼脂糖电泳显示二条区带,分别位于γ区靠边β区端和γ区中段。用DE_(52)离子交换层析纯化轻链。终产物经PAGE(用尿素和巯基乙醇解聚后)和SDS—PAGE分析均显示单一区带,分子量为26300Dalton。免疫双扩散检测K和λ均阳性,Fc阴性,表明系高度纯化的游离轻链(K和λ)。
The authors obtained a double-spread titer of K and λ chains from 1 urine of patients with double-light chain disease by ammonium sulfate salting-out (BJP). Agarose electrophoresis showed two bands, located in the γ region by the side of the β-region and the middle of γ region. The light chain was purified by DE_ (52) ion exchange chromatography. The final product showed a single band with PAGE (after depolymerization with urea and mercaptoethanol) and SDS-PAGE analysis with a molecular weight of 26300 Daltons. Immune double-diffusion assays were positive for both K and λ, Fc negative, indicating highly purified free light chains (K and λ).
