目的:研究晚期糖基化终产物(AGEs)诱导培养下,大黄酸对大鼠肾小球系膜细胞(RMC)表达转化生长因子β1(TGF-β1)和结缔组织生长因子(CTGF)的影响。方法:采用血清药理学及体外大鼠肾小球系膜细胞培养方法,用糖基化牛血清白蛋白(AGE-BSA)刺激肾小球系膜细胞,采用RT-PCR法检测系膜细胞TGF-β1mRNA、CTGFmRNA的表达。结果:晚期糖基化终产物上调TGF-β1mRNA、CTGFmRNA的表达,大黄酸呈剂量依赖性降低TGF-β1mRNA、CTGFmRNA表达,中剂量作用最明显。结论:大黄酸可通过下调AGE-BSA诱导的系膜细胞TGF-β1mRNA和CTGFmRNA表达,达到防治糖尿病肾病的作用。
AIM: To investigate the effects of rhein on the expression of transforming growth factor-β1 (TGF-β1) and connective tissue growth factor (CTGF) in glomerular mesangial cells (RMCs) induced by advanced glycation end products (AGEs) . Methods: Glomerular mesangial cells were stimulated with glycosylated bovine serum albumin (AGE-BSA) using serum pharmacology and culture of rat mesangial cells in vitro. The expressions of TGF-β1 -β1mRNA, CTGFmRNA expression. Results: Advanced glycation end products up-regulated the expression of TGF-β1mRNA and CTGFmRNA. Rhein decreased the expression of TGF-β1mRNA and CTGFmRNA in a dose-dependent manner, with the most obvious dose-effect. Conclusion: Rhein can prevent diabetic nephropathy by down-regulating the expression of TGF-β1mRNA and CTGFmRNA in mesangial cells induced by AGE-BSA.
