探讨超微铁皮石斛对脾虚便秘肠道细菌分子多样性的影响,可为其疗效和临床应用提供依据.制备小鼠脾虚便秘模型,设模型组、正常组、铁皮石斛传统汤剂治疗组和超微50%量汤剂治疗组,采集肠道内容物,提取肠道微生物宏基因组,通过细菌通用引物PCR扩增后用ARDRA分析其多样性.结果表明:正常组和铁皮石斛超微50%量汤剂组的OTUs(分类操作单元数)、Shannon指数和Brillouin指数分别相同,依次为6、2.5850和1.5820,模型组和铁皮石斛传统汤剂组的OTUs、Shannon指数和Brillouin指数也分别相同,依次为5、2.3219和1.3814,且前两组均大于后两组;铁皮石斛超微50%量汤剂组与正常组群落结构的相似性系数最大,为0.5000,模型组和传统汤剂组较小,为0.1818;聚类分析和主成分分析结果均显示,铁皮石斛超微50%量汤剂组小鼠肠道细菌分子多样性最接近正常组.因此,脾虚便秘造模使小鼠肠道细菌多样性遭到破坏,经铁皮石斛两种汤剂治疗,具有一定的调整作用,超微50%量汤剂效果优于传统汤剂.
To investigate the effect of Dendrobium officinale on the molecular diversity of gut bacteria in spleen asthenia constipation may provide a basis for its curative effect and clinical application.Mice spleen deficiency constipation model was established and the model group, normal group, Dendrobium candidum traditional decoction group and super Micro-50% decoction group, intestinal contents were collected, gut microbiomes were extracted, and the diversity of gut microbiomes was detected by ARDRA after PCR amplification by universal bacterial primers.The results showed that the 50% The OTUs, Shannon index and Brillouin index of the decoction group were respectively 6, 2.5850 and 1.5820, respectively. The OTUs, Shannon index and Brillouin index of the decoction group and the traditional decoction of Dendrobium candidum group were also the same, followed by 5,2.3219 and 1.3814, respectively, and the former two groups were larger than the latter two groups. The similarity coefficient of the population structure of Dendrobium candidum with 50% dextrose group and the normal group was the largest, which was 0.5000. The model group and the traditional decoction group were smaller , And 0.1818 respectively.The results of cluster analysis and principal component analysis showed that the gut microbiota in mice of Dendrobium candidum was the closest to the normal group.Consequently, Diverse Destruction, the Dendrobium Decoction of two kinds, a certain adjustment effect, an amount of 50% ultra better than the traditional decoction decoction.
