目的:用白芍总苷作用于腹主动脉结扎所致的大鼠心肌重构整体动物模型,观察白芍中苷类成分的抗心肌重构作用,分析其可能的作用机理,以期为其抗心肌重构的可能临床应用提供实验依据。方法:不完全结扎大鼠腹主动脉造成压力超负荷型心肌重构模型,假手术组动物除不进行结扎外其他操作与模型动物相同。造模一周后取存活的造模动物随机分为模型对照组、阳性对照药卡托普利45mg/(kg.d)组、白芍总苷100mg/(kg.d)组、白芍总苷200mg/(kg.d)组。各组动物灌胃给予受试药物或等体积蒸馏水20mL/(kg.d),每天给药1次,连续给药30天。末次给药动物禁食不禁水12h,进行如下指标的检测:颈总动脉插管测定血流动力学相关指标;称取体重、摘取心脏测定左心室指数及全心指数。结果:腹主动脉不完全结扎1个月,模型组动物的左心室指数、全心指数明显升高(P<0.01),收缩压、平均压明显升高(P<0.05)。与模型对照组相比,阳性药卡托普利45mg/(kg.d)可使左心室指数、全心指数明显降低(P<0.01),收缩压明显降低(P<0.05);白芍总苷100mg/(kg.d)可使左心室指数明显降低(P<0.05);白芍总苷200mg/(kg.d)可使左心室指数、全心指数明显降低,平均压降低(P<0.05)。结论:白芍总苷灌胃给药具有一定的对抗腹主动脉结扎所致心肌重构作用,其中的芍药苷可能是白芍总苷抗心肌重构的主要药效学成分。
OBJECTIVE: To observe the anti-myocardial remodeling effect of glycosides of Paeoniae Alba on the myocardial remodeling of rats induced by ligation of total paeoniflorin in abdominal aorta, and to analyze its possible mechanism, Myocardial remodeling may provide experimental evidence for clinical application. Methods: The model of pressure overload myocardial remodeling was induced by incomplete ligation of the abdominal aorta in rats. The animals in the sham operation group were the same as the model animals except that they were not ligated. One week later, the model animals were randomly divided into model control group, positive control group captopril 45mg / (kg.d) group, total glucosinolate 100mg / (kg.d) group, total glucosides of paeony 200mg / (kg.d) group. Each group of animals given intragastric test drug or equal volume of distilled water 20mL / (kg.d), administered once daily for 30 days. The last administration of fasting animals can not help but water 12h, the following indicators of detection: measurement of common carotid artery hemodynamics related indicators; weighed body weight, heart rate determination of left ventricular index and heart rate index. Results: Left atrium index and whole heart index of model animals increased significantly (P <0.01) and systolic and mean pressures increased significantly (P <0.05). Compared with the model control group, the positive drug captopril 45mg / (kg.d) can significantly reduce left ventricular index and heart rate index (P <0.01) and systolic blood pressure (P <0.05) Glycosides 100mg / (kg · d) can significantly reduce the left ventricular index (P <0.05); total glycosides of paeony glycosides 200mg / (kg.d) can make left ventricular index, heart rate index decreased significantly lower average pressure (P < 0.05). CONCLUSION: GABA has the effect of resisting myocardial remodeling induced by ligation of the abdominal aorta. Paeoniflorin may be the main pharmacodynamic component of total glucosides of paeony.
