[目的]分析2008年福建省手足口病患儿标本肠道病毒71型(EV71)分离株FJ08089全基因组序列特征。[方法]应用RD细胞,从患者疱疹液标本分离EV71,用RT-PCR方法扩增分离株病毒的全基因组,分析其基因组特征。[结果]FJ08089株的基因组长度为7 404 bp,其中包括5′端非编码区(5′UTR)长743 bp,病毒基因组编码区(ORF)全长6 576个核苷酸及3′端非编码区(3′UTR)长81 bp。ORF编码含2 192个氨基酸残基的多聚蛋白。[结论]FJ08089株基因组的结构与安徽地方株ANHUI703814十分相近,整个基因组的核苷酸同源性为93.8%,氨基酸同源性为92.6%,均属于C4亚型,而与Cox.A16国际标准株G10(U05876)差别较大,两者核苷酸同源性仅为77.9%,氨基酸同源性为84.0%。
[Objective] To analyze the whole genome sequence of FJ08089 isolated from enterovirus 71 (EV71) in children with hand-foot-mouth disease in Fujian Province in 2008. [Methods] RD cells were used to separate EV71 from herpes fluid samples of patients and the whole genome of the isolated virus was amplified by RT-PCR and its genomic characteristics were analyzed. [Result] The genome length of FJ08089 strain was 7 404 bp, including 743 bp in 5 ’untranslated region (5’UTR), 6 576 nucleotides in full length and 3’ end non - coding region in virus genome coding region (ORF) The coding region (3’UTR) is 81 bp in length. The ORF encodes a polyprotein with 2,192 amino acid residues. [Conclusion] The structure of FJ08089 genome was very similar to that of ANHUI703814 in Anhui Province. The nucleotide identity of the genome was 93.8% and the homology of amino acids was 92.6%, both of which belonged to the C4 subtype, but not to the Cox.A16 international standard Strain G10 (U05876) was quite different. The nucleotide homology between the two strains was only 77.9% and the amino acid homology was 84.0%.
