目的:研究大黄苷元对大鼠肝微粒体CYP450酶活性的影响。方法:将大鼠随机分组,对照组给予0.5%羧甲基纤维素钠(CMC-Na),诱导剂组苯巴比妥钠75.0mg/kg、地塞米松80.0mg/kg、β-萘黄酮75.0mg/kg,抑制剂组酮康唑50.0mg/kg,大黄苷元高、中、低剂量组分别为57.6mg/kg、28.8 mg/kg、14.4 mg/kg,计算各组肝微粒体肝脏指数,测定各组肝微粒体蛋白含量,用CO还原差示光谱法(Omura法)测定CYP450酶的含量,评价酶活性。结果:与CMC组及酮康唑组比较,大黄苷元CYP450酶活性升高;与苯巴比妥钠组、地塞米松组及β-萘黄酮组比较,CYP450酶活性低。结论:大黄苷元对大鼠肝微粒体CYP450酶的活性有诱导作用,且有随给药剂量的增加而增强的趋势。
Objective: To study the effect of rhubarb aglycone on the activity of CYP450 in rat liver microsomes. Methods: The rats were randomly divided into control group (0.5% CMC-Na), phenobarbital sodium (75.0 mg / kg) and dexamethasone (80.0 mg / kg) 75.0mg / kg, ketoconazole 50.0mg / kg, high, medium and low dose of rhubarb aglycone were 57.6mg / kg, 28.8 mg / kg and 14.4 mg / kg, Index, the content of liver microsome protein in each group was measured, and the content of CYP450 enzyme was measured by CO reduction differential spectrometry (Omura method) to evaluate the enzyme activity. Results: Compared with the CMC group and the ketoconazole group, the activity of CYP450 was increased. Compared with the phenobarbital sodium group, the dexamethasone group and the β-naphthoflavone group, the CYP450 activity was low. Conclusion: The rhubarb aglycone can induce the activity of CYP450 in rat liver microsome, and it has a tendency to increase with the increase of dosage.
