黄酮是黄芩的主要活性成分,具有抗肿瘤、抗炎和抗菌等药理作用。黄芩中黄酮成分数量多,通常成分含量较低,常规的质谱全扫描模式较难快速识别低含量的黄酮成分。母离子扫描-信息依赖-增强型子离子扫描(PREC-IDA-EPI)技术靶向提供母离子和碎片离子信息,降低了基质效应对低含量化合物检出的影响可快速准确地完成对化合物的识别。因此本研究在对黄芩研究相关文献进行了调研总结后,明析了黄芩中已发现的黄酮成分和可能存在的黄酮成分。然后基于母离子扫描-信息依赖-增强型子离子扫描(PREC-IDA-EPI)技术建立了针对黄芩中黄酮苷元和其黄酮氧苷的靶向UPLC-MS/MS分析方法,并将其应用于黄芩提取物中的黄酮成分分析。利用该方法共对黄芩中的97个黄酮成分进行了鉴定,包括29个苷元,68个氧苷。黄芩中鉴定的97个化合物为阐明黄芩药效物质基础提供了支持。基于PREC-IDA-EPI技术制定待测化合物的个体化检测方案可快速有效用于未知化合物结构鉴定。
Flavonoids are the main active ingredients of Scutellaria baicalensis Georgi, with anti-tumor, anti-inflammatory and antibacterial pharmacological effects. Scutellaria flavonoids in a large number, usually low in content, conventional mass spectrometry full scan mode is more difficult to quickly identify low levels of flavonoids. Then, a targeted UPLC-MS / MS method was developed for the determination of flavonoid aglycone and flavonoid glycosides in Scutellaria baicalensis based on PREC-IDA-EPI. Analysis of Flavonoids in Scutellariae Radix extract. A total of 97 flavonoids from Scutellaria baicalensis Georgi were identified by this method, including 29 glycosides and 68 oxoglycosides. The 97 compounds identified in Scutellaria baicalensis supported the elucidation of Scutellaria baicalensis based on the material basis. PREC-IDA-EPI-based technology to develop individual test compounds test program can be quickly and effectively used for unknown compound structure identification.
