Therapeutic biomacromolecules such as proteins and peptides have been gaining increased popularity to treat various diseases since 1970s(1).This significantly increases the demand of simple routes of delivering proteins and peptides via non-injection administration such as by oral, especially in chronic therapies. However, safe and effective oral delivery of protein and peptide therapeutics remains the biggest challenge, largely due to their unfavorable properties that result in rapid digestive degradation and low permeability through the intestinal epithelium. To successfully delivering proteins and peptides by the oral rout, many strategies attempt to provide structural protection of the proteins and peptide and improved absorption through the use of enzyme inhibitors, absorption enhancers, novel nanoparticle delivery system and chemical modification. However, each of these technologies has their limitations despite showing positive results.
