为研究珍稀食用菌大杯伞胞外多糖的抗氧化活性,采用菌丝液体发酵、乙醇沉淀获得胞外粗多糖,并利用DEAE 52-纤维素和Sephadex S-200凝胶柱层析对胞外多糖进行分离、纯化,以及抗氧化活性鉴定,结果表明:从胞外多糖分离到一种分子量均一的多糖CEP II,其抗氧化活性随着多糖质量浓度的增加而增加,清除自由基.OH,DPPH和.O2-的EC50值分别为0.119,0.185,0.215g/L;CEP II抗氧化活性显著,可有效去除羟基自由基、DPPH自由基、超氧自由基.
In order to study the antioxidant activity of extracellular polysaccharides of rare edible mushrooms, extracellular polysaccharides were obtained by mycelial liquid fermentation and ethanol precipitation. The extracellular polysaccharides were purified by DEAE 52-cellulose and Sephadex S-200 gel column chromatography The results showed that CEP II, a kind of polysaccharide with the same molecular weight, was isolated from exopolysaccharides and its antioxidant activity increased with the increase of polysaccharide concentration, scavenging free radicals.OH, The EC50 values of DPPH and .O2- were 0.119,0.185,0.215g / L, respectively. The antioxidant activity of CEP II was significant, which could effectively remove hydroxyl radical, DPPH radical and superoxide radical.
