目的:探讨青蒿琥酯和大蒜素联合作用对人骨肉瘤细胞株MG-63、U20S的体外抑制效果。方法:分别采用25、50、100μmol/L的大蒜素,10、20、40μmol/L的青蒿琥酯分别作用于体外培养的人骨肉瘤细胞株MG-63、U20S于24h。在显微镜下观察肿瘤细胞的凋亡情况,并采取MTT法检测实际的肿瘤抑制效果。以两者联合应用作为对照作用于人骨肉瘤细胞株MG-63、U20S,比较单独用药和联合用药的差异。结果:青蒿琥酯与大蒜素联合应用,对人骨肉瘤细胞株MG-63、U20S增殖有较好的抑制作用,并诱导其凋亡,较单独使用其中一种药物而言,联合用药效果更佳,差异具有统计学意义(P<0.05)。结论:青蒿琥酯与大蒜素在体外对骨肉瘤细胞株的抗癌作用具有协同效应,且两者的作用机理不同,值得深入研究。
Objective: To investigate the inhibitory effect of artesunate and allicin on human osteosarcoma cell lines MG-63 and U20S in vitro. Methods: Allo-osteosarcoma cell lines MG-63 and U20S were treated with 25, 50, 100μmol / L allicin and 10, 20 and 40μmol / L artesunate respectively for 24h. The apoptosis of tumor cells was observed under a microscope, and the actual tumor suppression effect was measured by MTT method. The combination of the two was used as a control in the human osteosarcoma cell lines MG-63 and U20S, and the differences between the single and the combination drugs were compared. Results: Artesunate combined with allicin had a good inhibitory effect on the proliferation of human osteosarcoma cell lines MG-63 and U20S and induced its apoptosis. Compared with the combination of artesunate and allicin, Good, the difference was statistically significant (P <0.05). CONCLUSION: Artesunate and allicin have a synergistic effect on the anti-cancer effect of osteosarcoma cell lines in vitro, and their mechanism of action is different and deserves further study.
