采用改进的小鼠肠系膜淋巴结(MLN)冰冻切片粘附结合实验法和MTT法,观察白血病患者尤其是B细胞性急性淋巴细胞性白血病(B—ALL)患者外周血单个核细胞(PBMC)体外粘附小鼠MLN高内皮小静脉(HEV)的活性变化及患者骨髓班单个核细胞(BMMC)体外粘附患者骨髓基质细胞(SC)的活性变化.结果:1.B—ALL,患者PBMC粘附HEV的阳性率和强阳性率均低于正常组;2.经抗淋巴细胞功能相关抗原一1(LFA—1)单克隆抗体(McAb)封闭后,B—ALL患者PBMC对HEV的粘附阳性率低于正常对照PBMC经同样处理组及自身PBMC末处理组。抗LFA一1McAab封闭也能使正常对照PBMC粘附阳性率和强阳性率下降;3.各型ALL患者BMMC与患者骨成SC的粘附率低于正常对照,各型患者组问没有显著差异。
The improved mouse mesenteric lymph node (MLN) cryosection adhesion assay and MTT assay were used to observe the in vitro adhesion of peripheral blood mononuclear cells (PBMCs) in leukemia patients, especially patients with B-cell acute lymphoblastic leukemia (B-ALL). Changes in activity of mouse MLN high endothelial venule (HEV) and bone marrow stromal cells (SC) activity in patients with bone marrow mononuclear cell (BMMC) adhesion in vitro. Results: 1. The positive rate and strong positive rate of PBMC-adherent HEV in patients with B-ALL were lower than those in the normal group; After being blocked by anti-lymphocyte function-associated antigen-1 (LFA-1) monoclonal antibody (McAb), the positive rate of PBMC adhesion to HEV in B-ALL patients was lower than that of normal control PBMCs treated with the same treatment group and their own PBMCs. . The anti-LFA-1 McAab blockade also decreased the positive rate and strong positive rate of normal control PBMC adhesion.3. The adhesion rates of BMMC and SC of patients with various types of ALL were lower than those of normal controls, and there was no significant difference among various types of patients.
