目的:观察软肝冲剂治疗前后肝硬化(LC)患者外周血单核细胞(PBMC)中转化生长因子(TGF-β1)、干扰素-γ(IFN-γ)和白介素18(IL-18)的mRNA表达及分泌水平的变化。探讨软肝冲剂对肝硬化治疗的作用机制。方法:RT-PCR检测20例正常对照组和25例肝硬化患者软肝冲剂治疗前、治疗2、4、6月后PBMC中TGF-β1、IFN-γ和IL-8的mRNA表达水平;ELISA法检测PHA体外诱生PBMC培养上清中TGF-β1、IFN-γ和IL-18分泌水平。结果:肝硬化患者PBMC中的TGF-β1 mRNA表达水平及TGF-β1分泌水平显著高于对照组(P<0.01)。而IFN-γ和IL-18的mRNA表达水平及IFN-γ和IL-18分泌水平显著低于对照组(P<0.01)。经软肝冲剂治疗后,TGF-β1 mRNA表达水平及分泌水平逐渐下降,6个月后与治疗前相比有显著差异(P<0.01),但仍高于对照组;IFN-γ和IL-18的mRNA表达水平及IFN-γ和IL-18分泌水平逐渐上升,6个月后与治疗前相比有显著差异(P<0.01),但仍低于对照组。结论:软肝冲剂能通过下调PBMC中TGF-β1 mRNA和上调IFN-γ和IL-8 mRNA表达,对肝硬化有较好的疗效。
Objective: To observe the changes of transforming growth factor (TGF-β1), interferon-γ (IFN-γ) and interleukin-18 (IL-18) in peripheral blood mononuclear cells (PBMC) from patients with liver cirrhosis (LC) before and after treatment with Ruangan Granules. Conclusion: Ruangan Granules have a good curative effect on liver cirrhosis by down-regulating TGF-β1 mRNA and up-regulating IFN-γ and IL-8 mRNA expression in PBMCs.
