目的探讨布地奈德对哮喘气道重塑大鼠原代培养的气道平滑肌细胞(ASMCs)上清液中白细胞介素13(IL-13)及干扰素γ(IFN-γ)含量的影响。方法 60只Wistar大鼠随机分为三组,每组20只。A组吸入卵清蛋白制作哮喘气道重塑模型,B组在A组基础上隔日1次吸入布地奈德气雾剂,C组吸入生理盐水作对照。8周后取材行苏木素-伊红(HE)染色,测量各组大鼠肺内支气管气道壁面积(Wat)/气道基膜周长(Pbm)及平滑肌层面积(Was)/Pbm;原代培养大鼠ASMCs,ELISA法检测各组ASMCs上清液中IL-13及IFN-γ含量。结果 A、B组Wat/Pbm及Was/Pbm均较C组增加,但B组较A组降低(P<0.05)。A组IL-13含量较C组增加,IFN-γ含量较C组降低(P<0.05);B组IL-13含量较A组降低,IFN-γ含量较A组增加(P<0.05)。结论布地奈德通过降低哮喘大鼠ASMCs间质中IL-13及增加IFN-γ的表达来减轻气道重塑。
Objective To investigate the effects of budesonide on the levels of interleukin-13 (IL-13) and interferon-γ (IFN-γ) in the supernatant of airway smooth muscle cells (ASMCs) cultured in airway remodeling of asthmatic rats. Methods Sixty Wistar rats were randomly divided into three groups of 20 rats each. Group A inhaled ovalbumin to make airway remodeling model of asthma. Group B received inhaled budesonide aerosol once every other day on the basis of group A, and saline as control. After 8 weeks, the samples were stained with hematoxylin and eosin (HE) to measure the bronchial airway wall area (Wat) / airway basement membrane thickness (PBm) and smooth muscle layer area (Was) / Pbm in the lungs of each group. The ASMCs were cultured on day 1, and the contents of IL-13 and IFN-γ in supernatant of ASMCs of each group were detected by ELISA. Results The Wat / Pbm and Was / Pbm in groups A and B were higher than those in group C, but were lower in group B than those in group A (P <0.05). The content of IL-13 in group A was higher than that in group C and the content of IFN-γ was lower in group A than that in group C (P <0.05). The content of IL-13 in group B was lower than that in group A and IFN- Conclusion Budesonide can reduce airway remodeling by decreasing the expression of IL-13 and increasing the expression of IFN-γ in interstitial of asthmatic rats.
