WST-1是一种新型的四唑盐。它生成水溶性的有色物质。比色时不需额外的溶解步骤。在本研究中我们将WST,1用于胃癌细胞株SGC-7901的抗癌药物筛选试验。我们的研究表明:①最适细胞接种数0.5×104-1×104/孔。②.WST-1显色法的最佳波长450nm。③.WST-1显色法重复性良好,组内变异系数为3.5%~8.7%。④WST-1法比MTT法敏感。⑤.WST-1法比MTT法简便。⑥.WST-1法用于抗癌药物筛选结果与MTT法具有良好的符合性。结果表明,WST-1法具有快速、简便的特点。是体外瘤株筛选抗癌新药和临床肿瘤药敏试验的可靠方法。
WST-1 is a novel tetrazolium salt. It produces water-soluble colored substances. No additional dissolution step is required for colorimetric comparison. In this study, we used WST, 1 for anticancer drug screening tests of gastric cancer cell line SGC-7901. Our study showed that: 1 The optimal cell inoculation number was 0.5×104-1×104/well. 2. The optimal wavelength of WST-1 coloring method is 450nm. 3. The WST-1 colorimetric method has good reproducibility with an intra-group coefficient of variation of 3.5% to 8.7%. The 4WST-1 method is more sensitive than the MTT method. 5. The WST-1 method is simpler than the MTT method. 6. The WST-1 method has good conformity with the MTT method for the screening of anticancer drugs. The results show that the WST-1 method is fast and simple. It is a reliable method for in vitro screening of tumor-bearing new drugs and clinical tumor susceptibility testing.
