目的探讨双黄连注射液对病毒性脑炎小鼠脑组织中NF-κB表达的影响。方法建立病毒性脑炎小鼠模型,给予不同剂量的双黄连注射液,分别于治疗后5、10、20d,通过Western blotting和RT-PCR,观察小鼠脑组织中NF-κB蛋白与mRNA的表达的变化。结果与对照组相比,模型组NF-κB蛋白和基因的表达水平明显增高(P<0.01),0.2和1.5mg/kg双黄连注射液治疗组中NF-κB蛋白和基因表达降低(P<0.05),而5mg/kg双黄连注射液可显著的抑制NF-κB蛋白和基因的表达水平(P<0.01)。结论双黄连注射液可以降低小鼠脑组织中NF-κB的表达水平,同时具有剂量和时间依赖性。
Objective To investigate the effect of Shuanghuanglian injection on the expression of NF-κB in brain tissue of mice with viral encephalitis. Methods The mouse model of viral encephalitis was established and Shuanghuanglian injection was given at different doses. The expression of NF-κB protein and mRNA in mouse brain tissue was observed by Western blotting and RT-PCR at 5, 10 and 20 days after treatment respectively. Change in expression. Results Compared with the control group, the expression level of NF-κB protein and gene in the model group increased significantly (P<0.01), and the expression of NF-κB protein and gene decreased in the 0.2 and 1.5 mg/kg Shuanghuanglian injection group (P< 0.05), and 5mg/kg Shuanghuanglian injection can significantly inhibit the expression of NF-κB protein and gene (P <0.01). Conclusion Shuanghuanglian injection can reduce the expression level of NF-κB in mouse brain, and it is dose- and time-dependent.
