目的观察不同年龄段大鼠脂肪间充质干细胞(adipose-derived stromal cells,ADSCs)生物学特点与年龄的关系,为临床寻找理想的种子细胞并迅速获取所需ADSCs提供实验依据。方法使用密度梯度离心法分离不同年龄段脂肪间充质干细胞进行培养,保留贴壁细胞传代,分析脂肪间充质干细胞的纯度,观察细胞生长情况,检测其增殖活性、细胞周期。结果不同年龄段ADSCs均贴壁生长,呈典型ADSCs形态和生长特征,传代细胞的增殖速度比原代细胞快,但随着年龄的增长,原代及传代培养ADSCs的增殖能力下降,生长周期延长。结论ADSCs的增殖能力和活性随着年龄的增长而降低,但各年龄段ADSCs均可满足临床不同患者组织工程种子细胞的要求。
Objective To investigate the relationship between the biological characteristics of adipose-derived stromal cells (ADSCs) and age in different age groups and to provide an experimental basis for finding the ideal seed cells clinically and obtaining desired ADSCs rapidly. Methods Adipose-derived mesenchymal stem cells (MSCs) of different ages were isolated by density gradient centrifugation. The passage of adherent cells was preserved. The purity of adipose-derived mesenchymal stem cells was analyzed. The proliferation and cell cycle were observed. Results ADSCs grew adherently at different ages and showed typical morphology and growth characteristics of ADSCs. The proliferation rate of ADSCs was faster than that of primary cells. However, the proliferation of primary and subculture ADSCs decreased with the increase of age, and the growth cycle was prolonged . Conclusions The proliferation and activity of ADSCs decrease with age, but ADSCs of all ages can meet the requirement of tissue engineering seed cells in different clinical patients.
