用人脐静脉内皮细胞体外培养方法和ABC免疫酶标技术,检测细胞表面抗原分布。结果表明:原代培养的10例人脐静脉内皮细胞,5例红细胞A抗原阳性;2例红细胞B抗原阳性;1例红细胞A、B抗原均阳性;2例红细胞A、B抗原均阴性。其中5例A型脐带血肉皮细胞分别检测HLA-ABC、HLA-DR、CD_4、CD_8抗原均阳性,自动图像分析证实有上述抗原分子表达。此外,本文研究证明,体外培养两周的内皮细胞具有表达抗原的功能。实验结果提示:体外短期培养的内皮细胞能保持其一般生物学和免疫学特征,其表达的抗原分子可能参与和调节着机体的免疫功能。
Human umbilical vein endothelial cells in vitro culture methods and ABC immune enzyme labeling technology to detect cell surface antigen distribution. The results showed that 10 cases of primary cultured human umbilical vein endothelial cells, 5 cases of erythrocyte A antigen positive; 2 cases of erythrocyte B antigen positive; 1 case of erythrocyte A and B antigen were positive; 2 cases of erythrocyte A and B antigen were negative. Among them, 5 cases of type A umbilical cord blood human skin cells were positive for HLA-ABC, HLA-DR, CD_4 and CD_8 antigen respectively. Automated image analysis confirmed the expression of these antigen molecules. In addition, this study demonstrated that endothelial cells cultured in vitro for two weeks have the function of expressing antigens. The experimental results suggest that endothelial cells cultured in vitro can maintain their general biological and immunological characteristics. The expressed antigen molecules may participate in and regulate the immune function of the body.
