目的探讨信号转导和转录激活因子3(STAT3)、血管内皮生长因子(VEGF)在儿童呼吸道复发性乳头状瘤(JORRP)组织中的表达及微血管密度测定(MVD)与JORRP复发、侵袭的关系,并且研究三者之间的相关性,以为JORRP的血管靶向治疗提供依据。方法采用免疫组化的方法,检测26例侵袭性JORRP组织及68例非侵袭性JORRP组织中STAT3、VEGF的表达,并用CD34标记血管内皮细胞,进行微血管密度计算。结果 JORRP侵袭组中STAT3、VEGF的表达阳性率高于非侵袭组(P<0.05),JORRP侵袭组的MVD值显著高于非侵袭组(P<0.01);JORRP组织中STAT3表达阳性和VEGF表达阳性的MVD值分别高于STAT3表达阴性和VEGF表达阴性(P<0.01)。JORRP组织中STAT3、VEGF的表达与CD34-MVD值呈明显正相关(分别为r=0.747,P<0.01;r=0.666,P<0.01),STAT3的表达与VEGF的表达也有显著的正相关性(r=0.464,P<0.01)。结论 JORRP侵袭组组织中STAT3、VEGF的高表达及高MVD值,表明STAT3可能是通过促进新生血管形成,参与JORRP的侵袭、复发过程。
Objective To investigate the expression of signal transducers and activators of transcription 3 (STAT3) and vascular endothelial growth factor (VEGF) in children with respiratory recurrent papilloma (JORRP) and the relationship between the microvessel density (MVD) and the relapse and invasion of JORRP , And to study the correlation between the three in order to provide a basis for the vascular targeting therapy of JORRP. Methods Immunohistochemical method was used to detect the expression of STAT3 and VEGF in 26 cases of invasive JORRP and 68 cases of non-invasive JORRP. The endothelial cells were labeled with CD34 to calculate the density of microvessels. Results The positive rates of STAT3 and VEGF in JORRP group were significantly higher than those in non-invasive group (P <0.05). The MVD in JORRP group was significantly higher than that in non-invasive group (P <0.01). The STAT3 expression and VEGF expression in JORRP tissues Positive MVD values were higher than those of STAT3 negative and VEGF negative (P <0.01). There was a significant positive correlation between the expression of STAT3 and VEGF and the CD34-MVD in JORRP tissues (r = 0.747, P <0.01, r = 0.666, P <0.01, respectively) (r = 0.464, P <0.01). Conclusion The high expression and high MVD of STAT3 and VEGF in JORRP-invasive tissues suggest that STAT3 may be involved in the invasion and relapse of JORRP by promoting neovascularization.
