Knv-Ⅰ是一种新近发现的抗癌基因,目前对其在白血病发病中的作用研究甚少。利用逆转录多聚酶链反应扩增Krev-ⅠcDNA片段,然后重组入质粒中而制备出Krev-ⅠcDNA探针,使用该探针进行Northern杂交检测Knv-ⅠmRNA表达水平,与正常骨髓细胞相比较,发现白血病细胞系HL-60、U-937中Krev-ⅠmRNA表达水平明显降低;14例急性髓系白血病中,1例表达与正常相当,2例轻度降低,9例明显降低,2例无Krev-ⅠmRNA表达。说明Krev-Ⅰ表达水平的降低在白血病的发生中可能起着重要的作用。
Knv-I is a recently discovered anti-oncogene, and its role in the pathogenesis of leukemia is currently limited. The Krev-I cDNA fragment was amplified by reverse transcription polymerase chain reaction and then recombined into a plasmid to prepare a Krev-I cDNA probe. The probe was used for Northern hybridization to detect the expression level of Knv-I mRNA, and compared with normal bone marrow cells, leukemia was found. The expression level of Krev-I mRNA in the cell lines HL-60 and U-937 was significantly reduced. In 14 cases of acute myeloid leukemia, 1 case was equivalent to normal, 2 cases were mildly decreased, 9 cases were significantly lower, and 2 cases had no Krev-I mRNA. expression. This indicates that the decrease of Krev-I expression level may play an important role in the occurrence of leukemia.
