不同结构的五甲川菁荧光染料常被用于近红外荧光探针,其灵敏度高,但光稳定性不理想.本文合成了一系列中位引入电子给体对氨基苯或对羟基苯的五甲川菁染料,研究了取代基对光稳定性的影响.实验表明,具有氨基和羟基电子给体的染料具有较高的光稳定性.飞秒瞬态吸收实验证明,氨基降低了菁染料分子的激发态寿命,光稳定性与激发态寿命成负相关,同时染料能较好地定位于活细胞中的线粒体.这对设计高耐光牢度的新型菁染料探针具有重要意义.
Different structures of pentachlorethrin fluorescent dyes are often used for near-infrared fluorescence probe, its high sensitivity, but the light stability is not ideal.In this paper, a series of intermediate introduction of electron donor p-aminobenzene or p-hydroxybenzene Wujiachuan Cyanine dyes, the effects of the substituents on the photostability were studied. The experiments show that the dyes with amino and hydroxyl electron donors have higher photostability. The transient absorption experiments show that the amino groups reduce the excitation of cyanine dye molecules The state lifetime, light stability and the lifetime of the excited state are negatively correlated, meanwhile, the dye can be located in the mitochondria of living cells well, which is of great significance for the design of new cyanine dye probes with high light fastness.
